Thomas De Raedt, PhD, is an Assistant Professor of Pediatrics (Oncology) in the Department of Pediatrics at the Perelman School of Medicine, University of Pennsylvania, affiliated with Children's Hospital Philadelphia. His research focuses on elucidating the mechanisms by which pediatric High Grade Gliomas develop and progress. His lab aims to understand crucial pathways involved in tumor development, their interactions, and how these insights can be exploited to develop novel therapies. To achieve this, his team molecularly analyzes human tumor samples, conducts cellular studies, and develops accurate in vitro and in vivo models. De Raedt's work particularly concentrates on pediatric High Grade Glioma (pHGG), a devastating disease with a median survival of approximately 12 months. His research emphasizes the role of the RAS pathway, especially in tumors driven by aberrant activation of this pathway, such as through loss of NF1. His lab investigates how mutations in epigenetic machinery co-occur with RAS pathway mutations and explores the cooperation between these genetic alterations. His team develops advanced in vivo models and tools to functionally identify and understand epigenetic drivers like ATRX, and how they cooperate with RAS pathway activation. Additionally, his research includes developing therapeutic strategies, including cellular and immunotherapies, such as cellular delivery systems derived from migratory interneuron precursors and Peptide-Centric Chimeric Antigen Receptors (PC-CARs) targeting NF1-associated tumors. His work also involves exploring new therapies through drug screening and innovative cellular therapies to improve outcomes for glioma patients.
