Jeffrey A. Johnson, PhD, is a Vilas Distinguished Achievement Professor and Director of the Bachelor of Science Pharmacology and Toxicology Program at the University of Wisconsin School of Pharmacy. His research focuses on the transcriptional control of neuroprotective genes and the prevention of neurodegeneration in diseases such as Parkinson's, Alzheimer's, Huntington's, and neuromuscular disorders. A central theme of his work is the investigation of dysfunctional proteostasis in disease progression and the development of viral-mediated delivery methods for protective genes to the brain. Dr. Johnson's laboratory has extensively studied the Keap1/Nrf2 pathway, which plays a critical role in mitigating mechanisms associated with Alzheimer's disease, including oxidative stress, mitochondrial dysfunction, inflammation, and protein aggregation. Activation of this pathway promotes the expression of detoxifying and antioxidant genes, enhancing cellular resistance to neurodegenerative processes. His research has demonstrated that Nrf2 activation in astrocytes confers neuroprotection both in vitro and in vivo, including significant attenuation of pathology in mouse models of Alzheimer's disease through reduced tau phosphorylation, neuronal loss, and amyloid-beta plaque load.
