  • Randomized Phase II and Biomarker Study of Pembrolizumab plus Bevacizumab versus Pembrolizumab Alone for Patients with Recurrent Glioblastoma

    Clinical Cancer Research · 2020 · 250 citations

PURPOSE: VEGF is upregulated in glioblastoma and may contribute to immunosuppression. We performed a phase II study of pembrolizumab alone or with bevacizumab in recurrent glioblastoma. PATIENTS AND METHODS: = 30). The primary endpoint was 6-month progression-free survival (PFS-6). Assessed biomarkers included evaluation of tumor programmed death-ligand 1 expression, tumor-infiltrating lymphocyte density, immune activation gene expression signature, and plasma cytokines. The neurologic assessmen…

  • Subtype switching in breast cancer brain metastases: a multicenter analysis

    Neuro-Oncology · 2020 · 111 citations

BACKGROUND: Breast cancer (BC) brain metastases (BM) can have discordant hormonal or human epidermal growth factor receptor 2 (HER2) expression compared with corresponding primary tumors. This study aimed to describe incidence, predictors, and survival outcomes of discordant receptors and associated subtype switching in BM. METHODS: BCBM patients seen at 4 tertiary institutions who had undergone BM resection or biopsy were included. Surgical pathology reports were retrospectively assessed to det…

  • Years of life lost due to central nervous system tumor subtypes in the United States

    Neuro-Oncology · 2025-06-10 · 7 citations

    articleOpen access

BACKGROUND: Years of life lost (YLL) is a disease burden measure quantifying the number of years lost due to premature mortality for a given disease. The present study sought to assess YLL for primary brain and other central nervous system (CNS) tumor histopathologies in the United States. METHODS: Mortality, incidence, and life expectancy data for mortalities occurring in 2018 were obtained from the National Vital Statistics System and the National Program of Cancer Registries. Tumor-specific Y…

  • Artificial intelligence for segmentation and classification in lumbar spinal stenosis: an overview of current methods

    European Spine Journal · 2025-01-30 · 6 citations

    reviewOpen access

PURPOSE: Lumbar spinal stenosis (LSS) is a frequently occurring condition defined by narrowing of the spinal or nerve root canal due to degenerative changes. Physicians use MRI scans to determine the severity of stenosis, occasionally complementing it with X-ray or CT scans during the diagnostic work-up. However, manual grading of stenosis is time-consuming and induces inter-reader variability as a standardized grading system is lacking. Machine Learning (ML) has the potential to aid physicians…

  • Non-recurrent mutations and copy number changes predominate pituitary adenoma genomes

    European Journal of Endocrinology · 2025-04-29 · 3 citations

    articleOpen access

OBJECTIVE: Pituitary adenomas (PAs) are common neoplasms. Our current understanding of the molecular basis of PA formation is incomplete. Routine implementation of targeted genomics has enabled the discovery of rare, potentially clinically actionable events. METHODS: We used a cancer-focused gene panel to sequence a cohort of 171 PAs from patients who underwent surgery at Brigham and Women's Hospital from 2012 to 2020. RESULTS: We identified known genetic variants enriched in specific PA subtype…
