• Body mass index and its association with clinical outcomes in acute liver failure

World Journal of Clinical Cases · 2025-10-11 · 2 citations

    articleOpen access

BACKGROUND: Acute liver failure (ALF) is a life-threatening multisystemic condition with high short-term mortality. With the growing prevalence of obesity and metabolic syndrome, it is important to investigate the clinical implications of high body mass index (BMI) on survival outcomes in ALF. AIM: To explore the impact of overweight and obesity on the clinical outcomes of patients with ALF. METHODS: A retrospective observational cohort study was conducted involving patients with ALF admitted to the Johns Hopkins Health System between January 1, 2000 and May 1, 2020. We performed Cox proportional hazards regression to identify outcomes, including the need for liver transplantation (LT) or all-cause mortality. RESULTS: = 0.008) than among survivors. Patients with overweight and obesity had a higher risk of all-cause mortality or need for LT by 2.22-fold (95%CI: 1.30-3.78) and 2.04-fold (95%CI: 1.29-3.39), respectively. Elevated BMI was associated with renal failure and higher grades of hepatic encephalopathy. Derangements in serologic markers, including alanine transaminase, lactate, and ammonia, were associated with a mortality risk or need for LT. CONCLUSION: In this large, retrospective study, with a diverse cohort of United States patients, Overweight and obese were independently associated with an increased risk of all-cause mortality or need for LT. This work highlights the importance of closely monitoring ALF patients who are overweight or obese for adverse complications and measures to improve outcomes in this vulnerable patient population.

  • Patchy nanoparticles by atomic stencilling

    Nature · 2025-10-15 · 15 citations

    articleOpen access1st author

Abstract Stencilling, in which patterns are created by painting over masks, has ubiquitous applications in art, architecture and manufacturing. Modern, top-down microfabrication methods have succeeded in reducing mask sizes to under 10 nm (refs. 1,2 ), enabling ever smaller microdevices as today’s fastest computer chips. Meanwhile, bottom-up masking using chemical bonds or physical interactions has remained largely unexplored, despite its advantages of low cost, solution-processability, scalability and high compatibility with complex, curved and three-dimensional (3D) surfaces 3,4 . Here we report atomic stencilling to make patchy nanoparticles (NPs), using surface-adsorbed iodide submonolayers to create the mask and ligand-mediated grafted polymers onto unmasked regions as ‘paint’. We use this approach to synthesize more than 20 different types of NP coated with polymer patches in high yield. Polymer scaling theory and molecular dynamics (MD) simulation show that stencilling, along with the interplay of enthalpic and entropic effects of polymers, generates patchy particle morphologies not reported previously. These polymer-patched NPs self-assemble into extended crystals owing to highly uniform patches, including different non-closely packed superlattices. We propose that atomic stencilling opens new avenues in patterning NPs and other substrates at the nanometre length scale, leading to precise control of their chemistry, reactivity and interactions for a wide range of applications, such as targeted delivery, catalysis, microelectronics, integrated metamaterials and tissue engineering 5–11 .

  • Machine Learning-Based Multiplex Immunofluorescence Staining from Immunohistochemistry with Generative Adversarial Networks

Journal of Student Research · 2025-02-28

    articleOpen access1st authorCorresponding

Immunohistochemistry (IHC) and Immunofluorescence (IF) are the two most commonly employed methods of cell staining, which aims to enhance visibility of cellular components for medical research and diagnosis. Immunofluorescence which uses fluorescently labeled antibodies to detect specific proteins is preferred over immunohistochemistry for multiplexing and colocalization which analyze protein distributions due to its transparent nature. However access is often barred due to high costs of reagents and equipment, slide quality degradation overtime, and technical limitations such as autofluorescence, and background staining from non-specific binding or cross reactivity antibodies. To address the aforementioned issue, we propose a novel machine learning-based immunofluorescence staining approach utilizing Generative Adversarial Network (GAN). The model intakes a digital pathology image of IHC and converts it to an IF image through feature maps. The proposed model achieved state-of-art performance with PSNR value of 30.67 and SSIM value of 0.8992. Additionally, the qualitative experimental results demonstrate the efficacy of the proposed method in enhancing generating high-quality IF stained images.

  • Origins of suppressed self-diffusion of nanoscale constituents of a complex liquid

    arXiv (Cornell University) · 2024-04-27 · 1 citations

    preprintOpen access

Understanding and ultimately controlling the transformations and properties of nanoscale systems, from proteins to synthetic nanomaterial assemblies, is limited by the inability to uncover their dynamics on their characteristic length and time scales. Here, we nevertheless demonstrate this ability using MHz X-ray photon correlation spectroscopy (XPCS) -- directly elucidating the characteristic microsecond-dynamics of density fluctuations of semiconductor nanocrystals (NCs), not only in a colloidal dispersion but also in a liquid phase consisting of densely packed, yet mobile, NCs with no long-range order. We find the wavevector-dependent fluctuation rates in the liquid phase are suppressed relative to those in the colloidal phase and relative to observations of densely packed repulsive particles. We show that the suppressed rates are due to a substantial decrease in the self-diffusion of NCs, which we attribute to explicit attractive interactions. Using coarse-grained simulations, we find that the extracted shape and strength of the interparticle potential explains the stability of the liquid phase, in contrast to the gelation observed via XPCS in many other charged colloidal systems. This work opens the door to elucidating fast, condensed phase dynamics in complex fluids and other nanoscale soft matter, such as densely packed proteins and non-equilibrium self-assembly processes, in addition to designing microscopic strategies to avert gelation.

  • Anion and Cation Migration at 2D/3D Halide Perovskite Interfaces

    ACS Energy Letters · 2024-05-14 · 32 citations

    articleOpen access

This study explores the ionic dynamics in 2D/3D perovskite solar cells, which are known for their improved efficiency and stability. The focus is on the impact of halide choice in 3D perovskites treated with phenethylammonium halide salts (PEAX, X = Br and I). Our findings reveal that light and heat drive ionic migration in these structures, with PEA+ species diffusing into the 3D film in PEABr-treated samples. Mixed-halide 3D perovskites show halide interdiffusion, with bromine migrating to the surface and iodine diffusing into the film. Cathodoluminescence microscopy reveals localized 2D phases on the 3D perovskite, which become more evenly distributed after thermal treatment. Both PEAX salts enhance the performance of photovoltaic devices. This improvement is attributed to the passivation capabilities of the salts themselves and their respective Ruddlesden−Popper (RP) phases. Annealed PEAI-treated devices show a better balance between efficiency and statistical distribution of photovoltaic parameters.

  • Peptide-Bound Glycative, AGE and Oxidative Modifications as Biomarkers for the Diagnosis of Alzheimer’s Disease—A Feasibility Study

    Biomedicines · 2024-09-19 · 1 citations

    articleOpen access

Background: The diagnosis of Alzheimer’s disease (AD) relies on core cerebrospinal fluid (CSF) biomarkers, amyloid beta (Aβ) and tau. As the brain is then already damaged, researchers still strive to discover earlier biomarkers of disease onset and the progression of AD. Glycation, advanced glycation end products (AGEs) and oxidative modifications on proteins in CSF mirror the underlying biological mechanisms that contribute to early AD pathology. However, analyzing free AGEs in the body fluids of AD patients has led to controversial results. Thus, this pilot study aimed to test the feasibility of detecting, identifying and quantifying differentially glycated, AGE or oxidatively modified peptides in CSF proteins of AD patients (n = 5) compared to a control group (n = 5). Methods: To this end, we utilized a data-dependent (DDA) nano liquid chromatography (LC) linear ion trap-Orbitrap tandem mass spectrometry (MS/MS) ) approach and database search that included over 30 glycative and oxidative modifications in four search nodes to analyze endogenous modifications on individual peptides. Furthermore, we quantified candidate peptide abundance using LC Quan. Results: We identified 299 sites of early and advanced glycation and 53 sites of oxidatively modified tryptophan. From those, we identified 17 promising candidates as putative biomarkers (receiver operating curve-area under the curve (ROC-AUC) > 0.8), albeit without statistical significance. Conclusions: The potential candidates with higher discrimination power showed correlations with established diagnostic markers, thus hinting toward the potential of those peptides as biomarkers.
